Mass Spectrometry è has become the best technology for verifying integrity oligonucleotes; each oligo is sold only if it passes the criteria based on MALDI- or ESI-TOF technologies.
Mass Spectrometry allows much more detection; sensitive to impurities like:

  • n-1 / n-x oligos
  • Purification
  • Incomplete deprotection
  • Acrylonitrile adducts
  • Identification of high concentrations of salts

Also è the most faster and more efficient for identifying potential MIX-UP products

Biofab uses two different types of mass spectrometry (MS) to better address the various quality issues; and quantity:

• MALDI-TOF è a technology with a “ throughput ” high; the limits of use of this technique are manifested if it is a question of analyzing long oligos or oligos with photo-labile modifications (for example the most common quenchers such as BHQs, DABCYL).

• ESI-TOF è less efficient in terms of throughput, but compensates perfectly as it è more suitable for long oligo analyzes and with photo-labile modifications

For better analysis of complex samples such as labeled oligonucleotides, Biofab also offers liquid chromatography (LC) coupled with electrospray ionization mass spectrometry (ESI-MS).
